What's the role of numerical aperture in microscope resolution?

The numerical aperture (NA) of a microscope plays a vital role in determining its resolving power, which in turn affects the detail and clarity of the images observed.

The numerical aperture is a dimensionless value that characterizes the range of angles over which a microscope can effectively gather or emit light. In simpler terms, it quantifies the microscope’s ability to collect light and resolve fine details of a specimen at a specified object distance.

Resolution refers to the smallest distance between two points that can still be perceived as distinct entities. The resolution of a microscope is directly influenced by its numerical aperture: a higher numerical aperture results in improved resolution. This enhancement occurs because a higher numerical aperture allows more light to enter the system, leading to brighter and more detailed images.

The numerical aperture is determined by two main factors: the medium between the specimen and the lens (known as the immersion medium) and the half-angle of the maximum cone of light that can enter or exit the lens. The formula used to calculate the numerical aperture is given by:

Here, nn represents the refractive index of the medium, and θ\theta is the half-angle of the maximum cone of light.

The refractive index of the immersion medium is crucial for enhancing resolution. For instance, oil immersion lenses employ oil between the specimen and the lens to increase the refractive index, which subsequently raises the numerical aperture and results in improved resolution.
