Staining is a widely used technique in microscopy that improves the contrast of microscopic images. In the fields of biology and medicine, stains and dyes are commonly employed to highlight structures within biological tissues, facilitating their observation, often with the assistance of various types of microscopes. Stains can be applied to examine bulk tissues—such as muscle fibers, connective tissue, and cellular components—or to classify cellular populations, as seen when differentiating types of blood cells. Additionally, staining can target organelles within individual cells.

The staining process typically involves treating biological tissues with a specific stain or a combination of stains that bind to particular tissue components or structures. The selection of a stain is dictated by the type of tissue and the specific structures intended for observation. For example, the Gram stain is used to categorize bacteria into two primary groups: Gram-positive and Gram-negative. Other stains, such as the hematoxylin and eosin (H&E) stain, are employed to highlight general features of cells and tissues.

Moreover, staining techniques can be utilized to detect the presence and distribution of specific chemicals or molecules within a sample. One notable method is immunohistochemistry, which uses antibodies that bind to specific proteins. These antibodies can be labeled with a stain or a fluorescent dye, enabling visualization of the protein’s location under a microscope.
