What is a protease and how does it work?

A protease is an enzyme that catalyzes the breakdown of proteins into smaller peptides or individual amino acids.

Proteases play a critical role in various biological processes by facilitating the hydrolysis of peptide bonds in proteins. They are essential for functions such as digestion, blood clotting, and the immune response. Proteases can be categorized based on their catalytic mechanisms into four main types: serine, cysteine, aspartic, and metalloproteases.

For instance, serine proteases contain a serine residue in their active site, which directly attacks the peptide bond to cleave the protein. In contrast, cysteine proteases utilize a cysteine residue to perform the hydrolysis. Aspartic proteases feature two aspartic acid residues that work in concert to facilitate protein breakdown, while metalloproteases require a metal ion, such as zinc, to assist in the catalytic process.

The activity of proteases is tightly regulated through various mechanisms. These include inhibition by specific inhibitors, activation by other enzymes, and changes in environmental conditions such as pH or temperature. Dysregulation of protease activity can contribute to a range of diseases, including cancer, Alzheimer’s disease, and cystic fibrosis.
